Yes, but also no.
Long answer:
Overall resistance values can be reduced to -100% (double damage) with the default ruleset. However, it will never be displayed properly if a resistance is below 0%. At present, the only resistance that can be brought below 0% is fire resistance, as a handful of effects apply a penalty to it.
Rune smiths debuff enemy resistance, so it should work too i guess.
