You're pretty much correct on the story quests. However, they're also a major source of reputation and you need that in order to recruit NPCs to your village. If memory serves, there were one or two mandatory quests just to get started but after that, you're free to do your own thing. There is an announcement board in the main town (name escapes me yet again) that has quests you can do for points, along with side quests by NPCs from the other villages.
As far as location...good question. I'm not convinced I'm happy with my first spot (is anybody ever?) but I'm going really slowly on this one and have but a few buildings, so it'll be easy to move when I've found "it." Hopefully others will offer their suggestions because I'm curious where people are building.
Oh, and make sure to check out the abandoned buildings. They offer a lot of loot to get you started. Beware, the animals have all been pumped with steroids. Even at 10% strength, they can be hard to kill if you're a poor hunter like I am. Thank you unlimited HP for letting me play at all in The Oxbow! Oh, and if you're one who prefers to turn off bandits, that's not an option. Of course, if you're a skilled hunter, the new found animal strength might give you the change you've been looking for.

-1 or 2 mandatory quest hm. Well on day 1 of release I did do quests until it pointed me to the notice board. But I stopped there as the game needed and had its patches.
Assuming the notice board and NPC side quests are optional, I might skip them, unless they offer decent rewards.
-In the first game/map I always made a "starter" base, than moved after year 1
-I'm hoping this island I'm going to check out, will be what I want.
-Enemies have more str... cool, I better avoid them until I get the good stuff
