In this case to acess the estus soup you must hit the "E" key on your keyboard which is located toward the top right of your movement keys ("W", "A", "S", and "D").

Should You Actually Use The Estus Soup?

You should without a doubt use the soup unless you are a collector since using the estus may decrease it's value because it no longer exists (at least at the location you consumed it at).
The estus soup should not be doubted. It's powers are far greater than mine, yours and maybe even Patches when he's in onion knight form. When all out of estus flasks, poisoned, and at 1 HP you are able to get a quick boost of HP and get straight back into praising the sun. However, this is not the correct way to soup it up!

The Correct Way To Get Value From Estus Soup

Estus soup is very scarce, so correct usage is the key to victory. Using it after you have ran out of estus flasks is a big mistake. When you are out of estus flasks the chances you will die goes up since you are not able to replenish your health. This means you should not use estus soup when out of estus flasks. Estus soup tends to be in areas that are covered with enemies and if they are not then the parts shortly after will be. This only adds to why you need to have remaining estus flasks to make full use of estus soup; your chances of death go up in areas as dangerous as the estus soup locations.

If you have been paying attention to all of the information you will now realise that you must use the soup when you have estus remaining in order to make sure the soup is valuable and does not go to waste. Just remember, it may be worth dying instead of drinking the estus soup purely because soup is a finite resource in DARK SOULS™ III and you may not be satisfied with the value you received from it if you die after using the soup.
