Creating a DDS file is as simple as saving whatever you're editing in a DDS format (using my guide to get the right settings).
All you need to do to start creating is click File then New... , and choose your resolution.
You can make it any resolution you want (even different aspect ratios) as the game should just stretch them to fit the object. A common example of a texture that doesn't use a 1:1 ratio is the hands - They are often made to be twice the height, because they are wrapped around the fingers, and need more detail in less space.
The easiest way to make textures for Skyrim is to make a copy of an original texture, and then edit that (resizing it before you start) - using the original image as a guide layer that you can delete later.
