First things first: Introduction! (I didn't have one, so you already owe me...)
Let's get the obvious one out of the way: The Game demands your second player to have a Controller. Either you hook up one of dem smelly X-Box controllers or you use a Playstation one running on an emulator. My personal choice was DS3 Tool ( just google it, I got it from my friend on a stick ) and then setting that to the X-Box compatibility settings.

Once you are in-game it might prompt you to set all buttons individually, which is easy to do. Just follow the on-screen instructions that appear after the logo screens and set the controls up the way you/your buddy like them.

After that, you can choose to either set up a new game that starts in co-op, or continue with an already existing save that spawns your buddy right next to you at your (in-game) uncles house.
Secondly: Controlling yourself! (Seriously, don't panic! I said DON'T! Damnit, too late.)
Alright. Now that I assume that you both are in the game we can get started with the basics. The safest thing for you, most likely the keyboard player would be to sit in a corner while you and your buddy are safe and let your new-found companion try all the buttons. ALL of them. See what works, what doesn't.

For me, the Layout presented itself as following:

Prompt: A Button

Sprint: X Button

Movement: Left Stick

Weapon selection (If avaivable): Right Stick

Draw/holser selected gun: Right triggerpad

♥♥♥♥/shoot gun: Right Trigger

Journal: Button
The Good, the Bad and the Guide Section: Friendly Fire and other quirks of the game
So, that little weapons testing session didn't go too well and you lost a couple of hats? Damn. I should have told you before, but there IS friendly fire in this game, and I'll tell you how to efficiently counter-act it in a pinch. But here are some other observations I have taken note of.

To Player 1:

You drive the Story. You are the only of you two that can interact with stuff, and therefore you are moreso in control then your mate might be. You open doors, buy things and revive your partner when he/she goes down. But the game will "end" if you die, so make sure you keep your butt out of danger.

And To Player 2:

As a companion you make sure that your Bro/Sis travels well. You provide gunfire, draw enemies to your quasi-immortal ass and support them with everything that they might be doing. You cannot interact with quest givers and shop items, but with hats and money on the ground as well as collectibles. Be supporting or ♥♥♥♥ around, just make sure it's all in check with your partner. I do not encourage violence, though that controller goes well as a bludgeoning device should your plus-one act irresponsibly ( no really, stop hitting him/her already!) .
Cover my Four! Tactical Combat in Westerado and avoiding Friendly Fire.
Rule number one on this part of the list: Don't shoot eachother!

"But how?" You say, exhaling frantically as another bullet whizzes past your brand new ten-gallon hat, your friend giggling as you take cover in fear.

That's mighty easy: Go tactical about teh combat thing 'n' stuff (Professionalism and guide writing don't go hand in hand...). As all weapons in the game shoot only on the x-axis, you can easily divide the screen into a top and a bottom 50-50 style. This will drastically reduce the rate of accidents, especially in the stagecoach/buffalo herd missions.

Another good method for avoiding peppering your fancy headgear is adjusting your kit. If your friend is carrying the shotgun they are in no great danger of blasting you to bits as long as you, responsible player, do not engage the same cluster of enemies. Or you just take the rifle and stand next to each other while blasting foes from a distance.

Or you can encircle the enemies if you are really skilled with revolvers. And remember to stop shooting eachother if all foes are suffuciently hatless and beaten.
Five makes a Coyote: Environmental Hazards
Time to relax for Player 2! Since only Player one can go to Areas and Player two will be automatically teleported through the power of Whiskey and their fancy Poncho, they will have to face the perils of the desert all alone. You as the Co-op partner can just sit back and watch them getting a stingy toe-to-toe with scorpions.

Or come with them just for the sake of companionship. Your call.
Six-Shooters: Conclusion.
If you have endured my bossy Instructioning for this long, you are, at least from my point of view, ready for the perils, adventures and riches of the wild frontier. Swig that last shot, kick that tumbleweed across the street and go over to the Station to meet your new Gunslinger Partner. And to you, Blue Bandana Man: Welcome aboard.

This is just me (Mr. Blue Bandana) hangin' out with mah Sis. Mah real Sis.
