Ninja Gaiden games can't or at least couldn't use Reshade on their own because it would crash the games. Special K allowed people to bypass that issue by loading Reshade through Special K, which didn't crash the game when setup correctly.
This guide was written for that very specific case and right now links to pretty old versions of both Special K and Reshade. Your best bet is to just get the new version of Reshade and follow the instructions there and forget about Special K. If it's Special K that you specifically want then you should look at the wiki or the discord server for help. The discord server is pretty active.
It's best to use a reshade version older than 5.0 if following this guide.

I don't see that button in the reshade install.
You can google around Special K see discussions about it online.
The link is pretty outdated right now, but works with the guide and build of the game.
One issues might be that you did the [API.Hook] Fix for NG3. You don't need to do that. Infact you shouldn't do it. Just try to reverting everything under the [API.Hook] back to true.
