For the missions, unless they specifically state it must be done in one mission, can be done over the course of the entire run so you can slowly complete the use 5 items over time.
As for advice I'd recommend keeping your deck as small as possible, and keeping your deck to the Attack(red) and Defend(blue) cards for your first run to make it easier to pull off the combos. IMO you should likely skip the card they give you at the end of battle unless it works well with what you have already or it will only hurt you by slowing down your deck.

Seriously though, eriia's comment is right on point. Deck management is key. If you have the opportunity to burn a card, take it, unless it's tied to a fight you know you probably won't win (like an early high elite). I usually only add cards if they REALLY support my build, or if they are upgraded gamebreaking legendaries. Most of the early card drops are a hard pass. You can usually clear the first boss with the starter deck on non Transcended, especially if you pick your elite battles wisely.
Different missions, like the use 5 items you mentioned, can be difficult if you don't have the cash, skills, or cards. Some skills, like Smuggling, make one item in the store free for the rest of the game. Cards like Reservoir will let you use an item once without consuming it. Given the circumstances, use 5 items can be a breeze or nearly impossible, depending upon the temper of the Random Number Gods. This notion can extend to almost any mission requirements.
Final note, don't be discouraged by others who say you can beat the game in 1-3 hours. These are most likely Slay The Spire (STS) fans, of which I'm most definitely one, or folks who have been playing card based roguelikes for years. If you have a lot of experience with those kind of games, then you'll adapt very quickly to this game. Neoverse is basically STS with no map and more booty. If not, don't sweat it, you'll learn each character's meta in time, and you'll be throwing down T-10 runs in no time.
To be honest I've never liked these types of games - (STS Roguelike) though I really want to. I truly believe this is the first game genre that I truly do not understand the mechanics of.
I watched a live stream of this and there were like lions and massive skill unlocks and crazy frantic animations - all of which I have not yet to experience.
I do enjoy it though as it's very casual, but trophies and etc just seems like a massive grind to obtain stuff.

How to get MOAR trophies for the shinies? What does "end score will not be ranked' mean anyway? Is there a leaderboard?
I agree about this being more casual, but there's more challenge to be had on higher Transcended levels, Hunter mode, and daily decks. Yes, there's several ladders. You can see them from the Rank menu. I don't know why you're getting the "end score will not be ranked" error. Are you playing offline or have Steam Cloud Sync turned off?
Concerning end game and grind, that's really all there is with roguelikes. For many players, including me, the challenge is making the most of what you can with what the RNG gives you. One typically plays a game like this for the sake of the gameplay, rather than completion. There's also ladders, achievements, and skimpy outfits to unlock if you're into all those kind of things.
Thank you for your tips. I am still working on understanding shuffle and I still make some mistakes, but I've been learning a lot now about Claire specifically with the vampire build start and her skills as well as chosen cards and such.
Just last night I finally discovered how the combo system works! I thought you had to play - attack/defend/infinite/constant in order to proc the combo as per the instruction menu and was wondering why it wasn't working. Turns out, that little bar is telling you what to play! Once I got started on that (I'm color blind - so purple and blue sometimes miss) I was just slashing away at the levels not even paying attention to what card I was using, just the colors haha!~
I'm getting the score not ranked due to starting out with gear equipped for + skill points - ALSO another reason I wasn't getting combos and parry. Once learned, I quickly unequipped and am now trying to get enough for the mask that inc % on trophies.
Unsure a out outfits and confused at character start where it states "#Attempts _____unlock" with then an XP bar.
