There are several ways, some more legitimate than others.
Having allowed visitors makes it a bit harder since you don't know if it's one of your citizens or not.
For your own dwarves, looking at someone that wants to drink but hasn't met that need for a long time usually makes it much easier, since undead don't eat or drink.
This also often results in vampires and necromancers having a harder time maintaining a good mood (eating, drinking and sleeping are frequent good thoughts for living dwarves).
The usual legitimate way to figure out who the vampire is is to do it through the justice system, interrogating dwarves, trying to figure it out, but it can take a while before you have enough clues to make a proper guess.
