As for your specific questions, I don't really know for sure since we aren't shown numbers or anything, but I'm pretty sure it doesn't include mutations that are not active. I would assume it uses whatever rank of whatever mutations you have active. I don't believe the damage the droplet itself adds is affected by the number of active mutations - did you see something that made you think it does?
If you don't yet have phase 3 of lil fist, I have found that if the killing blow isn't from the fist attack itself, it's not counted towards lil fist. So don't equip things that do extra hits like assassin (bleed dot), brood helm (poison dot), or truffle tussle (explosion counts as club/generic damage). The explosion and damage from DoT ticks are not counted as fist attacks, so if that's the damage the kills the enemy, it doesn't get counted towards lil fist. Disable those mutations and farm red lawn mites until get phase 3.
edit: don't think whittle wizard has any effect with unarmed unless using one of the candy accessories, but then wouldn't be using power droplet. If using power droplet with fists, would not be doing any minty/sour/spicy damage for whittle wizard to affect. Power droplet lets you use effects even if they're for other WEAPONS. Whittle wizard isn't for any specific weapon.
