Without water, 2-3-0 village + lots of heavy DPS tower or DPS + stalls like 0-0-5 Ice Monkey. DDTs have camo, lead and black properties so 2-3 village lets all monkeys in radius hit camo, destroy all types of bloons and attack faster. DDTs are fast so something like 0-0-5 Ice Monkey slows them down so the rest of your towers can burn them down. Or if you have enough DPS, just burn them down.
x3x Village is fine too.
this bomb shooter can fully stun lock all ddts, well actually it stun locks everything except for BADs and bosses
the lower path mermaid is also amazing against ddts, especially when paired with any damage-over-time effects (like glue, fire, bleed)
but since you were asking for heroes... I dont think many heroes do much against ddts tbh
etienne allows all towers on the map to see camo, so any of your regular lead popping towers become a DDT defense
high level Sauda can bleed the DDTs, Ezili can attach her spirit stuff to DDTs but neither is enough to destroy them i think
Usually Bloon Sabotage, Jungle Drum, and MIB are common answer.s But things such as dealing with fortified MOABs on R95, could be a much bigger threat than DDT.

I choose Sulfur Spring and only use the middle lake water and edge as example.
I can get:
- Twin Gun Sub as opener
- Gwen as main Hero that buffs Fire Wizard, but also gives fire attack to other towers
- 4-0-1 Sub as cleanup + decamo,
- 0-4-2 Wizard as main DPS, for early-mid game
During the period to round 80, I have 3 main issues:
- MOAB (Use a single MOAB Assassin)
- Rushes (Use Maelstorm)
- Lack of buffs (Abyssal Warrior for pierce buffs, 3-0-0 Alchemist for Phoenix)
Those towers are not randomly bought. They do something to Super Ceramics, DDT, and buffed MOABs. Embrittlement and Glue Storm are great addition to ability heavy based setup that features lot of projectile.
With this I can get basically any tier 5, but Super Maelstorm is enough to carry the game because the map has loop, and Gwen can consistently trigger Heat It Up at right spot.
So understanding what you really need is better than buying "best" towers. There are many other options you can try.
Then it's just a matter of cashing out your investments to buy enough versatile dakka. If all else fails MIB makes all dakka versatile, but there are towers that don't require it that can be sufficient for a while.
