How to upgrade to 5-5-5 tower?

As soon as I choose 2 paths, the third one gets locked and I have no idea how to make it work!

You can't. You can get 5 upgrades on one path, 2 on another, and 0 on the other. Choosing which upgrades to get is part of the strategy.

...Or can you? Maybe if you get enough xp for your dart monkeys, boomerang throwers, ninja monkey, or monkey buccaneers, you could get three tier 5s, then merge them into one 5-5-5 tower! Try it!

Basically, yes. It has always been like this:
There are 2 paths. On one, you can go above 2 and the other, you can't.
For 6, it's always been like this:
You have 3 paths. You can choose 2 of them. One you can upgrade to 5, the other one to 2. Only ONE tower that follows a specific path (5 on one) gets to keep the 5. If there are more that want to have the 5, you can't. Only one dart monkey can have a 5 on the path you choose and the rest can only go up to 4. Paragons are for a few monkeys and they let you sacrifice all monkeys of that kind to make an ultra-powerful one. If you want to play COOP, you can add me.

A single tower can't have more than one path on tier 5 in a legit match. If you have seen it anywhere, it is mod or something like that. You can only upgrade 2 of the 3 path. One of them to tier 5 and another to tier 2.
But there exists 2 special things for some towers:
Paragons, williamatics16, told it. If you have unlock this abiltiy for dartmonkeys, ninjas, boomerangs or ships, you can use this special ability. For this you have to build from this sort (as example the dart monkey) one tower with tier 5 in first path, another tower with tier 5 in second path, a third tower with tier 5 in third path. Then you can sacrify them with a lot of money and a paragon (very strong tower) arises. This could be called a tier 6 tower, but right name is Paragon tower.
The other special exception is the vengefull true sun god: You can build on special way something like the paragon tower with the supermonkey. This is called "the vengefull true sun god". It is not a paragon, just similar. It is the strongest tower in game.
If you want have more Information about a tower or something the bloons wiki (google it) is very usefull.
