What does "Playthrough" mean?
I've just finished Destroyer and proceeded to T-bone junction to play DLC.
When I restarted the game, save data makes me choose between playthrough 1 and 2
What's different?

Enemies tougher. They begins on 36 lvl. Also more legendary stuff.

Playthrough 2 is like " New game + ", a game mode that lots of games have. You play the game all over again ( Playthrough 2 still has Quests and side quests ) with tougher enemies but you will also have access to more powerful weapons, with higher stats. Legendaries are more common as well.

After you finish Playthrough 2 ( it ends when you kill the Destroyer, same as in Playthrough 1 ), you will unlock Playthrough 2.5. This Playthrough will not have Quests and/or side quests tough. Enemies will be even more tougher than in Playthrough 2 but they will drop even more powerful stuff, and of course, Legendaries are " very " common, in this Playthrough.

You might wanna have a read, in Borderlands Wiki, to better understand what's " Playthrough " and how each of them work.

Edit to add

In every new Playthrough, you keep all your lvls and stuff ( weps, shields, class mods, etc ), from the Playthrough you just played before.

Also you pretty much need to go on playthrough 2 in order to fully level up anyway.
I hate to say this to the OP, but you just ♥♥♥♥♥♥ up your own gameplay, and here's the reason why.....

when you kill the Destroyer, and decide to travel to any of the DLC areas and complete the missions there, those Claptrap Rescue Missions will become glitched and they won't reward you with anything.

So if anything, you should've played up till the Eridian Promotory, and travel to a DLC map then. That way, you can complete the Claptrap Rescue Missions without glitching them out.

no its just on playthrough 2 you cant beat the game until u get the sdu's from the claptraps in the dlcs (execpt knoxx thats very rare to get at anytime) playthough 1 will always reward a sdu for every mission (except knoxx like i mentioned before thats very rare to get one playthough 1 or 2)

Do you mean missions which reward SDU(free 3 slots)?
I've completed Knoxx dlc by killing knoxx and looting his armory(not crawmerax. I'm 41)
and I was rewarded with a grenade mod in CL4P-TP rescue mission in Locked Palace.
Is it glitched?
BTW if penalty given to me is only limited size of backpack I don't mind lol

the only time it glitches is when you kill the Destroyer, before you even have a chance to play through the DLC's (regardless of playthrough)

if your interested in the backpack sdu's and how to get them all (72) then go on wiki it explains it very well just make sure you play the dlcs to get the sdu's before you beat the game on playthrough 2 that also means just entering the the vault because that will put you in "2.5 playthrough" . a good tip for when you farm the first 5 claptraps on playthrough 2 (those are the only claptraps that have a chance of rewarding them) is to instead of alt f4 out of the game is to just kill yourself, bleed out, and while your teleporting save and quit youll see a message about how the game cant save whhile your teleporting its a fast way to farm for anything.
