Moe and Marley - I suggest leaving them until a bit later, and do not go after them when you first get the missions. They can be a pain, since they are different elemental types. If you can, try to take them on singly. Both of them attacking you at once is really not much fun.
Personally, at the start of each of my characters I rather like the clipper - 9 Toes' incendiary pistol. It may be rated fairly low, but it does work well. For later on in the missions, there is Krom's pistol as well. Very, very useful I have found.
Skags - learn to shoot them in the mouth when they stop and howl at you. That is their critical point.
2nd playthrough does start in Fyrestone again, but this time you got all your guns and ammo and shields. Great. But all the enemies are now stronger.
This is true of the third playthrough (2.5). Bonehead at level 72 - not that easy to put down. :)
Fatflape - I sympathise with running out of ammo on that tentacled monstrocity. Been there, done that.
As far as weapons are concerned. I have played B1 a lot, including spending a long time farming for good weaponry - Knoxx's armoury has been looted many, many times, so I have a full bank and backpack of good quality weapons, all the way up to Lvl 69 pearlescents and legendaries. More than I can sensibly use.
If you want, tell me what specifically you are looking for, the level you are, and if I have something suitable, I can drop in to your game and drop some goodies for you to try out. If you just want to bump up your bank balance - I have got guns whose price is listed at $?,???,???. (over a million) No more worries about running out of money to buy ammo :)
Of course, some battles are plain unsuitable for that playstyle - notably, Skagzilla.
Oh, and about gear again - "Tediore is a way to go", as said by Marcus =) All Tediore oranges regen your ammo, so you can either use them as is, or bring with you and swap them in some safe place or before major battle.
No problem. The offer is still there for some <whatever> weapons if you want them.
Go elemental, and as many x4 you can carry. Explosive is generally the most important to have - not many are resisatnt. Corrosive and incediary carry on working for you, and shock kills shields very quickly and stuns opponentsfor a bit.
Have fun. It is a great game.
Not sure what class is your char but if you play with Roland ( Soldier ), the Support Gunner class mod is very handy ( a must have/use imo ) because of the Regen Ammo feature, if you run out of ammo too quickly.
Things on PL 2 will get easier. Enemies will be stronger but more powerful weps ( and shields, class mods, etc ) will come easier to get as well... and you will know already all the enemy/boss "tricks" and how to beat them =)
" on a 2nd playthough, does that mean are weapons or content carry over? "
All your lvls, weps, etc will carry over to PL 2 ( and from PL 2 to PL 2.5 )
Sadly, you can't import Borderlands weapons into Borderlands 2. This makes me sad. :(
yes me too, i hate that they didn't include the Pestilent Defiler in BL2
@Dathip feel free to ask me for help any time you need, that's what friends are for :)

Glorius Serpens and Pearl Chimera for me - both awesome weapons. So is the defiler :)
But on the bright side, they do have Sledge's shotgun and the Jacobs Skullsmasher in B2. There may be more, but those are two I have dicovered so far. The Skullsmasher in B1 is Nasty. I have seen the result of a headshot leave the lower torso and legs. Overkill somewhat. (No such thing - hehe)
I personally find the Bessie (a Jakobs pearlescent sniper) to be the best in the game, due to it being 100% accurate when zoomed in, and it's more nastier than the Skullmasher. plus, I find both the Serpens and the Ajax's Spear to be the best machine guns in the game, as the latter weapon will sometimes spawn with the Ogre Explosive accessory.
the HellFire, hands down, is the best SMG in the game, due to how the shots proc on every hit and quickly drains an enemy's heath (the same goes for the Firehawk pistol). the version I use with my Mordecai character is a real powerhouse, as I modified it further to deal extra shotgun damage (meaning it deals almost 700-Damage per hit), on top of its 12.5 fire-rate.
