the Science is based on how far ahead or behind you are in science to that civ.
Now see where your available caravan is located currently. If it's not in the city you like to have as departure point, just move the caravan there. When you're in the right city, click on the button that creates a caravan, and choose the desired destination city from that list.
PS: when browsing the list of available trade routes in the overview, check the icon on the left that shows the type of caravan that can be established. If it shows the icon of a boat and you only have camel caravans, you won't be able to establish that trade route, because it needs a trading ship.
Science and faith do not get the x2 sea multiplier, but hammers and food do. But then hammers do not scale with city bonuses, so all of the trade calculations seem to be all special and weird. A sea route typically gives +25% production to your capital. So one way to go with Venice is a few overseas routes while your capital cranks out the wonders.
Trade routes do not update. They are much like a static agreement what expire after 30 turns.
