I found this out when I gave my PC "The Three Finger Salute" Ctrl-Alt-Del
Doing this will bring up the Windows Task Manager and under the Processes Tab you should see it as a running prog.
You'll need to close that and then you'll be able to run the game with no problem.
Right click on it in the Task Manager and "End Task".
This is because the Civ V exe file also runs the Civ V SDK. [/i]
There should be no need to reboot your PC or verify your game cache - although this won't hurt if you do so.
If it happens again give this a go, it worked fine for me.
