Assisted = Get another hobby
When you can only kill a few zombies, because you want the best time, and you have to pass a hall filled with zombies it's a thrill. Screams and bloody arms come at you from all angles. You know you're one mistake away from death, and if you don't dodge this next zombie just right, the three behind you will catch up.
It gets the heart racing, and really is a blast.
If you decide to do more than one play through, i say start on standard. The unlimited saves, auto saves, and slightly easier zombies, will let you explore the game at a better pace. You can experiment with weapons, and methods, and save as much as you need to. After that you can do hardcore and get a more challenging experience, and a bit more fun.

but you answered your own question if no auto saves are a deal breaker for you ;)
i can tell you though that the game always gives you an ink ribbon before a boss fight, so you shouldnt have to spend too much time on backtracking
edit: oh and just to point it out, "hardcore" makes it sound alot harder than it actually is ... if the game had difficulties like hard/very hard/nightmare its around "hard"
I guess I'm getting lazier at worst or less masochistic at best, because for this game I started off on Normal and then switched to Hardcore for the next run. But honestly Hardcore would have been perfectly manageable for a first run, just make sure not to run out of ink ribbons (save every time you reach a new typewriter or solve a "puzzle").
Reason why i will play just once is difficult to explain, but i plan to play it again on hard and achievements run in ~2 years.
