You can syringe him or poison the cake, or poison what he drinks. You don't need it twice anyway for the challenge

Just FYI but those can also be done in contracts mode, they are not target specific, any of the versatile assassin challenges do not require main mission targets

The behaviour in Miami has change in the last patch to make people now use trash cans instead of toilets

Maybe Bangkok has the same bug.

You can get it easily with coins. There's no need for poison they just need to be standing near a toilet. There's plenty of people who have paths near toilets for contracts and even both main mission targets have triggers that put them near enough to distract them the rest of the way
