How to get Kharaa Sample

I can't get Kharaa Sample because i don't have injection ampoule. Where can i get it?

Assuming you mean the antidote, it's in one of the small caves that you need a spy pengling to access. The location is circled in red on the map you find in Phi Robotics. Once you have that antidote, insert it in the droid machine on the top level near the leviathan.

im stuck in the fabule and i dont know what i must to do

I had the antidote on me, but the on screen prompt said I couldn't do anything without the injection ampule. Is there an in between step needed?

You need to insert the antidote into the small robot (next to the frozen leviathan) with the injection syringe. The robot will do the rest.
