Since there is just vague information around the internet regarding this Mesuking card, I'm going to post this in case it helps someone in the future.

According to the guides you need to gain victory in 20 Mesuking battles to trigger the Professor giving you this card. This is absolutely not 20 victories total, i well surpassed that given the substories and the majima battles, it's 20 outside of the substories. at least it was for me. and I had to go outside and come back in and speak to Professor to get him to give me the card after I had surpassed 20.

Also as an aside, using a weaker card against Masashi actually makes the fight easier. after about 7 tries and losing, I started trying random cards, using the western hercules beatle I just spammed rock the entire time and easily won. I tested it 3 more times and it was the same outcome. Maybe it's just RNG but i thought i'd share.

Remember rules:
1) Never make first step-AI is cheater and always counter you if he play after you.
2) Don't be predictable- if you win with scissors, for example, don't try it again.
Follow this and you beat every opponent easily enough.

You can easily farm this by re-fighting Tomohiro since he always picks rock
