I could take a guess that the snake in the canyon (that we kill later on) is male ,
The snake in the cave guarding the shrine was female, due to being very protective in her nature to protect her nest

One in the shrine, one at the bottom of the bridge/valley or whatever thats called.
The one in the shrine doesn't have injury on his eyes iirc

After killing the snake , later on i when't back to the shrine cave as i missed some items ,
the snake was still guarding the shrine ...if i recall it correctly
