As for run modifiers, I honestly don't remember. The original announcement said "It is not required to beat the game to use these modes - you can turn on a setting in the options to use them." Maybe it has changed since. Or maybe it just doesn't get offered right when you start a new game until you beat the game for the first time.
Have a fun time!
The alternate game modes are hidden by default and normally show up after you beat the game once. However you can also enable them early in the "gameplay" tab of the options on the main menu. Scroll to the bottom and toggle "Advanced Game Modes" to On.
Here's a screenshot of the menu option in question:

Though do be aware that enabling the alternate play modes may result in your first playthrough being much more difficult than normal. Monster Sanctuary is a mechanically dense game and the alternate game modes kinda assume you already understand how the game works. Which is why they're "intended" to be used after beating the game once.
Don't be afraid to start a new file without any modifiers if you find the other modes to be too spicy.
This is a game where the Player will make progress likely for different reasons.
One game mode removes monsters that may have been gained in previous runs.
In order to enjoy the game when playing modes that randomize, lock monster starting teams, or punish losing monsters in battles (none of these are required however can think of these as different pieces of a hard-core run) then it's good to have extra saves (there's 90 save files to keep progress).
So if the Player does something and wants to change what happened. That extra save file will be very beneficial to have.
And if the Player chooses to only enable "Relics of Chaos" (that adds more content in the form of item-equips that carry weaknesses with power) and using that to help farm the Arena later then that is also great to have on another save file.
I always keep 2 saves of any Run for convenience.

The Forgotten World can also be accessed before the "Final Story Area" which means that the Monsters gained there can be used later in the Abandoned Tower's Boss Fights. Provided that the Player collected a Monster that has the required Traversal abilities to overcome the terrain to reach the Forgotten World.
That also means on Randomizer Mode that the Player could access Forgotten World even earlier than normal if the correct Monster is encountered early.
There are restrictions on New Game Plus using late-game monster terrain abilities (kept from previous runs) early-game before encountering them during exploration.

The "Forgotten World" and the more recent "Relics of Chaos" are the freely (already present - no extra downloads needed) added expansions that offer new content.
And are accessible in-game each according to their unique "unlock" methods.

There's also this to remember when the Player encounters New_Game_Plus:
