They have a limited range (1.8 km by road?), which you can check by mousing over the vehicles in the hospital's menu.

check that the road that goes from the hospital to the Main road is actually build.
Maybe its waiting to be build and the construction office did not deliver something
It can also be a road somewhere els that was not build or that you upgrade that was never build. So if the ambulance have no "route" to the people they need to pickup then they wont drive anywhere.
Check that your hospital have electricity.

When I check the hospital the roads are yellow

The yellow road is what the ambulances can reach.
Same way you can check fire station, construction office, snow plow station, and so forth.

I recently observed at my current game that the trucks at the woodcutter were not been maintenanced. I checked if the trucks from the mechanic garage can reach it.
Turned out they just couldn't.
I can ether build another garage or build a shortcut.
I did the shortcut.
Now it works.

To come back to the hospital.
The yellow road is just for the emergency.
When the ambulance won't go out maybe there is just no emergency at this moment.
So imo that's a good thing.
