Once you build an end station, add it to your bus line (preferably before the buses pick up their passengers, so that people don't wait in the end stations). For best performance, put the end station near to the next stop on the line. Then, click on the end station and you can set the spacing to the bus line: either leave it on 'Automatic Spacing' where the time between bus departures is calculated by the game, or go for 'Manual Spacing' where you set how many seconds there will be between bus departures.
The alternative (which isn't as good, but doesn't need you to build an end station) is to select the bus line and check the box for "Enable Spacing" or some term like that. After your buses run a few cycles of the line's route, this option will make them try to keep an equal spacing between vehicles. However, unlike the use of an end station as described above, this option has the buses maintain spacing by *slowing down* if they get too close to the vehicle ahead of them, which can artificially increase travel time for passengers on that bus route.
