I'm not sure if the prima macula carrier dying is the end. It seems like one person is chosen from a particular bloodline to hold this power. They also mention a part of lore that says something like "digging deep down will reveal the stone" probably the philosopher stone which is tied deeply to alchemy itself. These are mysteries that may only be answered in the sequel.
To give a more definitive answer though. I'd say that no, the death of the carrier doesn't necessarily mean game over for Earth. The main villain gains his powers by taking Hugo's blood which gives him equal powers. It would be possible to take Hugo's blood and make virtually anyone a carrier as long as they survive the process of hitting the threshold. Maybe if the carrier were to die without this happening it would mean the end but as long as the power can be given, or even taken for that matter, there is a way to still stop the end.
