Try launching through the EXE as the launcher has issues for some people.

Same problem here..

Just run the main file "BorderlandsGOTY" in the main folder. You can turn off the Launcher anyway, in the game Options.

That'll work too! Can make a shortcut to Desktop as well. The Launcher is kinda irrelevant because all of it's option(s) are in the games' options anyway.

Looks like the game isn't saving properly. Not sure if this is due to the Launcher issue.

I was so excited to replay this game, but I've been having the same issue and have tried fixing it for 2 days. I'm just done, I'll wait until the devs decide to patch out some of the million bugs people have been having with just launching and saving.

try to go to this directory "steamapps\common\BorderlandsGOTYEnhanced\Binaries\Win64" and change the name of the "BorderlandsGOTY" file to "Launcher" and the name of the original "Launcher" file to "BorderlandsGOTY". It worked for me just fine. Game runs without any issues, it saves normally.

Same problem here. In task manager I could see the .exe running, then go to 'not responding', then close. I tried everything here with no luck. Messed with Nvidia stuff with no luck. Went to eventvwr and found the following

Faulting application name: BorderlandsGOTY.exe, version: 1.5.0.0, time stamp: 0x5c995f45
Faulting module name: EZFRD64.DLL, version: 0.0.0.0, time stamp: 0x51932b88
Exception code: 0xc0000005
Fault offset: 0x0000000000002383
Faulting process id: 0x1c64
Faulting application start time: 0x01d4f2f241ca3ad7
Faulting application path: C:\Program Files (x86)\Steam\steamapps\common\BorderlandsGOTYEnhanced\Binaries\Win64\BorderlandsGOTY.exe
Faulting module path: C:\Windows\USB_Vibration\00791800\EZFRD64.DLL
Report Id: ed7b30e1-1267-4844-a70f-f0cbf74e5bd6
Faulting package full name:
Faulting package-relative application ID:

The Faulting module path stuck out to me for some reason, so I went there and renamed that .DLL to .DLL.bak. Game started after that. Not sure if it'll keep running or not, but after a couple hours of frustration, I thought I'd post my progress.

Data Execution Prevention is the most common cause of 0xc0000005 (memory read: ACCESS DENIED).

For Windows 10:
Start > Exploit Protection > Program Settings > Click the '+' left of 'Add Program to Customize > Add by Program Name > Type 'borderlands.exe' > Click 'Add'

WARNING: Adding by 'Choose exact file path' is more secure.
WARNING: This makes you more vulnerable to hacking if the game code is compromsied.

Find any setting that is turned 'On' and check the box left of 'Override system settings', then click the button that says 'On', and it will toggle to 'Off'.

For Windows XP, Vista, 7, 8, 8.1:
Start > Run > 'sysdm.cpl' > 'Advanced' (Tab) > Under 'Performance' click the 'Settings' button > 'Data Execution Prevention' (Tab) > :

There are two settings:
Turn on DEP for essential Windows programs and services only
Turn on DEP for all programs and services except those I specify

The second is more secure (assuming this is selected) click 'Add' > navigate to 'borderlands.exe' > click 'Open'

Disabling DEP via Exploit Protection, and installing the vcredist_64.exe, along with NVIDIA Physx fixed the issue for me.

OR

change the setting to
Turn on DEP for essential Windows programs and services only

OR

Start > Type 'cmd.exe', Right click, and select 'Run as Administrator' > Type 'bcdedit.exe /set {current} nx AlwaysOff'

WARNING: This will disable DEP for the whole computer, and is not recomended from a security standpoint.

If these don't work then it is possible you have DEP enabled forcibly via either the EMET, or via Group Policy.

Doing the above, and installing vcredist_64.exe (Visual C Redistributable), along with PhysX_8.10.29_SystemSoftware.exe (NVIDIA PhysX) from the prerequisites folder solved my problem.
