Which mission is Ptolemaios 2 Kai WS in?
How do you unlock it?

If I remember correctly it pops up in all missions from the 00 Movie and there's no other way to unlock it but to fill the get-gauge.

If you want to fill the Get gauge really fast, play on Extra. It takes between 5 to 7 kills to fill the bar up for any character, even ships. And as a bonus, the Guest units are buffed on Extra, so they can mostly solo the stage on their own.
