You will get that option as soon as you are able to have mandates.
It will say "increase trade route with X" and give an indicator for the costs. Influence, worked stone, bread, boards and tools for the respective trade routes.
Every level up of the trade route will increase the amount by 125. You can only upgrade the trade routes if you hit the trade max (Just doesnt show the option).
If you look at the trade route page it labels each trade route with the level and amount of goods they can trade to you. There is also a box thingy showing how many goods you have to trade with them in order to be eligible for a trade route upgrade.
Then after you are eligible for an upgrade to the trade route it will show a decisions by the Bailiff/Courtier/Emissary you can take to upgrade the route. It will cost you that amount of goods or influence to upgrade.
If you are pursing aspiration then some of those routes will not be allowed because you are not allowed to build a Monastery or Castle.
You can also take Bulk Trade which allows most trade volume, but prices slightly worse.
With Bulk Trade the trade level amounts are:
Level 1 23
Level 2 45
Level 3 113
Level 4 180
