i dont have 500 rep gang mission at night club and i have mission that says work your way up to the local gang i started over twice

yup you need to work your way up to unlock downtown and to unlock the pharmacy to get the stuff you need to work for the cartel/gang

So i'm already gang rep 1 with La Bellena but it has given me the quest to work my way up to one of the gangs which I clearly have already done?

ok i found the other way but it still says i have to retrieve a package that ive already recieved and also how do i work my way up to local gangs?

Did you already started gang stuff before that mission poped-up? If yes, then you were too
fast unlocking districts. "work your way up" should trigger before any gang contacts you.
