The easiest but most costly is to use the Awake items. These items when used in the gift menu for Mistles allows you to instantly master the gift at the cost of a certain number of them as well as Haze. There's four types of Awake: MJ018, MJ109, MJ212, and MJ310. You can grind these in the depths fighting bosses and sometimes Knights in the Memory of Player section. The MJ018 is the most common, its usually just a random drop with enemies. Using Revenant's Greed will help you find most of them, but the best is figuring out which bosses in the depths drop the specific Awakening you need to master the gifts you want.
For reference sake, I'm currently lvl 165.
Once you unlock the Tower of Trials, equip all of the gifts you wish to master alongside Murasame's Blood Code, then head into Tower of Trials III.
Open up the first floor, hop down and kill the big monster. Look at how much your gifts have skilled up. It seems each time you kill this monster, your mastery goes up maybe 15-20%. Not an accurate number, but that's how much of the bar it seems to fill. Once you kill the guy, teleport back to your mistle, rinse-repeat.
Depending on your skill level this fight may be a big challenge, but it definitely helps skilling up decently quick. If you're really good, then just keep going down the tower, but if that particular monster gives you troubles, then I'd recommend just farm him.
If you don't have Murasame's blood code yet, then just keep going through the story. This will help you skill up everything else before you hop into NG+.
In end game, it's faster to farm depths bosses for Awake MJ items using the one-hit kill build.
No, you only need to get the keys once. You keep them for the entire playthrough, so you don't have to worry about getting them every time. After you defeat the main boss the first time, rest at the Mistle and you can immediately go and fight it again.
That and the Tower of Trails and Murasame’s gift to master things faster. These are effective ways to level up.
