there are a few instances in the game where you make such checks hidden, for example at the start of some dialogues. so sometimes you wont know that you have an option like this simply because it's not there (because you failed the check at the start of the dialogue) as opposed to normal skill checks where you roll for the check after you picked the dialogue option.
Stag Lord gets nausea, basically removing him from the fight for a few rounds which is very helpful.
The poisoned wine? It's in Thorn Ford, where you rescue Jhod. There's a tent you can loot that has a nature skill check in the camp he's in. If you fail the skill check, there's no text to tell you though. You just fail it.
You can get inside the fort later on though, yes. There are a variety of options to make the stag lord encounter easier there.
