There is a specific route that leads to a spot where one of the floor tiles has a visible special symbol on it (easy to not notice if not looking for it), and that's the direction you have to go in to avoid the giants room (vs. going the "wrong way" from that same spot).
I honestly don't remember exact details.
I do remember that once you've been there/finished it once, that large room with all the initial starter floor tiles are all available at once and you can choose whichever one you want to use first. Meaning you could enter the main bosses room in one try, every time, if you knew what the correct original elevator tile pattern was.
But like I said, when you do it first time that isn't available - I think they want to force you to be aware of that red giant room because it's important post-game. That said I'm pretty certain you can avoid repeating the red giant room more than once. Maybe there's a random factor to it tho so sometimes you could and sometimes not. Dunno.
Edit: my memory is the correct floor tile to take is one where if you trigger it but don't stand on the elevator, as you watch the platform go down you'll see an activation circle on one of the walls vs. on the floor of the elevator tile. That was the one that had the symbol.
If you stand on the elevator as it goes down, it's too late to get off/out by the time you realize it's not the right one. It was something of that sort anyway.
