The boss would always be on stage 6 unless you were to loop around to the beginning.
On stage 5 if you interact with the side plates on the teleporter they will close, then if you use start the teleporter event, when it's done you would loop back to stage 1 but it would be stage 6.
You can also loop by using the blue portal and gold portals I believe.
And you can certainly loop using the Bulwark's Ambry portal on stage 5.
As far as I know you can infinitely loop.
You can obliterate on every stage 3 through the celestial portal after looping to end your run if you don't wish to fight the boss. Obliterating also rewards you with Lunar Coins that you can use in future runs.
Basically it's how many stages you can see before you start getting repeats.
Stage 1 is always one of four stages (two environments)
Stage 2 is always one of two stages (two environments)
Stage 3 is always one of two stages (two environments)
Stage 4 is always one of two stages (two environments)
Stage 5 is always Purple.
After that you'll 'loop' back to Stage 1 (assuming you didn't go through to the final boss) and get one of the four stages. So, 5 stages in a loop. The Celestial Portal always appears on stage 3 after the first loop, and the boss' teleporter is always gonna be on stage 5.
