You can also try fire breath potion trick if extra tentacles are removed when you return to z0001.
I am spiking bud (which I tend to forget about and derp out) which will get me past 1000 and hopefully with 9 champs fielded I can get enough bud to clear 1600.

In a variant that remove some slots, spiking BUD means going to area 1 or so and set a full party, hit a good BUD and go on with FB potion until the degradation stops progression again, rinse & repeat.
You can also spike a BUD in other situations, like going a few tens areas before & stay there the time some building buffs, like Astarion's Well Fed, have the time to stack up. With a few building up abilities, you can commonly gain 100 areas with that method. Or even more in some situations (cumulating sticky debuffs & build up abilities... but that starts to be longer and more boring to set up and tune up).
yes e425 is threshold to beat z1600, everytime i can get to that, no problem finishing.
No, only in variant when slots are removed during the course of the adventure. In most case, z1600 (or higher) can be reach without doing anything special after initial set up as long as you've the gear & legendaries.
As kab_reg added, in most case, I only check if my BUD is above e425 after initial set up (or will grow up to this point if I use champions like Evelyn or Blooshi), sometimes it's tricky with strong restrictions, other times I know I'll end with a e800+ BUD without efforts.
