It plays whenever a sound file whice is supposed to be played is missing, you should try to verify your game files, it will then download anything that is missing.
To do this, right click on the portal 2 "icon" in your steam library and choose properties, after this, select the "Local Files" tab and click on "Verify intergity of game cache" after that, just let steam load and when its done, it should say that some files are missing and that they are being downloaded, if this dosent work, just tell me here!
By the way, that sound is from Half-life 2!
the sound effect (error.wav) is the sound equivalent of the glowing red "ERROR" model, when the game tries to play a sound effect that it doesn't have, it plays the fiddlesticks sound instead. i imagine its there so developers immediately know something is wrong since its pretty hard to miss.
