Depends on the dialect and localization you're using.
Where I'm from, most ending vowels followed by S will devolve into schwas.
So it becomes phonetically congruent to "glad us", which is the same way we would pronounce the name Gladys.

I've always just pronounced it as "Glah-dose", considering the capitalization in the name. Like Gla-DOS, rather than something like Glad-OS.

nevermind, Harry101UK`s voice actors pronounce it like Glad Ohs or maybe he just does i dont think he can voice all the cores, wheatley and defective and normal turret
