A bit yes and no

A basic understanding of the SAO universe can stop you being puzzled by a few things and help explain who people are and how they act

But the game for the most part does hand hold you at the beginning to try and catch you up on things and explain in the quickest way possible who the companions are.

But hey, if you know nothing about SAO then you can enjoy Yuuki to her full potential

SAO - dude is op in a mmo in which you will die irl if you're killed in game. guy gets a gf, he has a forced harem which he ignores and saves world.

Asuna - Kiritos IRL girlfriend and mother of Yui

Sinon - A player of an MMO FPS called Gun Gale Online. In these games she was drawn into Sword Art Online due to a technical glitch

Leafa - Kiritos IRL sister although technically his cousin. She ended up like Sinon and got trapped in SAO due to a glitch

Philia - A player Kirito met in the Hollow zone in Sword Art Online. She was trapped in an area of the game not meant to be accessible to other players and as such for a while thought she was an AI

Seven - A child prodigy who Kirito met in Lost Song. She is doing research on Virtual Reality

Yui - An AI Kirito and Asuna found in Sword Art Online who they ended up adopting

No. The game has it's own original story and at the beginning of the game you will get a recap of who every character is and the events that have unfolded during the game's universe and the original Anime/Light Novel story.
