How to pick multiple sidekicks?

You can pick FROM multiple sidekicks. Once you make your choice of sidekick and leave the tutorial town, you will have that sidekick for the rest of the game. There are three to choose from, a doctor, a rancher and a prospector. You will find them around the first town and as I recall they each take a small sidequest to unlock them for use as companions.

Oh I thought you can have like 2 sidekicks at a time.....

No, just the one, but I found that was enough. I've only used Alice the Doctor so far but she can heal you, her attack is a shotgun, it's weak but it hits all enemies, and she has a saw that can one hit kill skeletons.

How to get her?

If you talk to the townspeople one of them will tell you she likes Nurse Whiskey. After you find that out you can find a bottle.....in the cellar of the pub I think? Go to her door and give it to her. Then you have to go through a conversation tree and convincer her to accompany you so that she can get revenge.

You can get the whiskey first, but you'll still need to talk to the Hostler to find out she likes that brand anyway.

You actually can get two sidekicks.

I am not gonna spoil how, but it involves the buffalo pile.

You can also get one of the people that you have to kill in the tutorial town as a sidekick, But since i don't want to spoil anything, You'll have to find out

Seems a bit odd to me that there would only be one...

By "extra" sidekick, I meant in addition to one of the four pardners.

Bill acts more like a summoned creature.
