That having been said, a strong case could likely be made for either the ancient rune weapons of the dwarfs (The first ones, such as Gotrek's axe) or other, similarly fated weapons for empowering their users and granting immortality, which is a powerful bolstering force in and of itself.
The sword of Khaine is canonically also pretty crazy.
However, I think if we're fully honest with ourselves, the most powerful weapon in the lore would likely be a steam tank or a cannon, unless you which to stretch the definition of 'weapon' to include mounts. As there is a very real, very solid reason for swords to have been replaced with longer range emplacements, and the fact is, ogres carry cannons as -guns-

I would bet my money on Widowmaker though.
I always hear Widowmaker. In the end times, doesn't Tyrion essentially pull an Aenarion and basically go into hulk mode due to it? Or it was cursed after Aenarion I don't remember I just know it is some God-weapon that even transforms based on the users perception or something. I could google this but so much less fun.
Edit: Wait, does it make them the avatar of Khaine? He's the same God of War type in fantasy as in 40k right? Idk, just always odd to me as I view him as a type of chaotic-neutral that seems to be able to wipe his ass with Daemon's. Well until his space-race got raped. I really have no idea as to his role in fantasy other than probably angry and war.
The Sword of Khaine appears differently to whoever perceives it. Aenarion and Tyrion had swords, Malekith saw a septer as it would give him the power to rule like he wanted, I think Tethlis the Slayer saw a spear, or was it Caledor the Conqueror?
But yes, I'd say Widowmaker is strongest as it cursed an entire bloodline just from being drawn and, depending on the source, may have cursed the Elven race itself by giving some members a terrible bloodlust.

Yet it is not the greatest weapon in WFB. The greatest weapon is weapon of Krakanrok the Black. Why? He is bigger than a mountain. Even Kholek looks puny next to him. Try to survive a hit from that.
