Is the 500kg bomb 1 use only?

Your plane will rearm when its out of all ammo across its strategems.

You can manually rearm earlier if desired.

No, it's the same as the other eagle stratagems, once all your charges are depleted it will reload after a ~2m30 cooldown

eagle strategems can replenish during missions. takes like 2min 30sec to rearm which will do automatically if you use up all their ammo or if you manually call it to rearm after you use 1 or more of its uses

No it doesn't unless its your only eagle strategem.

I have all hangar upgrades, you get 2 Eagle 500 before needing to rearm. If you dropped your 2 eagle 500 bombs and have other Eagle drops in your loadout, plane won't rearm unless you order it, If it's the last Eagle drop you have charges for it will rearm auto. Eagle rearm with upgrades takes a bit under 2 mins and will recharge all of your eagle stratagems.

Best thing about eagle, with full charges you have a lot of up front damage esp if you have 2 or 3 eagle stratagems. With upgrade you can order back to back eagle strike in around 10 secs. So long as you have charges you can continue to rain down fire where orbitals take longer.

Downside to eagle is preparation, eagle stratagems are also linked, can't be used in a combo attack with another of your eagle drops and require more thought to their use.

Example being if you used both eagle 500s and want to rearm, all your other eagle stratagems go on cooldown, this can become a big problem. Been caught few times with pants down having a couple of eagle cluster left and forgetting to rearm the 500s, usually doesn't end well.
