Contrary to most of the other comments, fire damage is actually the slowest way to kill a tank, dealing only 100 damage per second.
While it is the safest method to light him on fire and run, OP asked for the fastest way, so I've compiled a list of how quickly every weapon can kill tanks with the stats from here.
Keep in mind that being good at killing tanks does not necessarily mean these weapons are the overall best in the game. After all, tanks aren't the most prevalent nor biggest threat.
It's also important to keep in mind that these are the best possible scenarios with no external influences - So if a weapon is said to kill a tank in 5 seconds, that means it will kill a tank in at least 5 seconds under the best possible condition of all players being able to skip reloads and landing all their shots. In actual gameplay this will likely be 10-15 seconds instead, since you won't always be right next to a weapon spawn, nor land all your shots, and other random factors like not always having a full mag and your teammate's performance are going to have a significant effect as well.
For example, in an average team that actually shoots at the tank you can expect it to take about twice as long to kill, whereas in a pub lobby where everyone runs around like headless chickens you can multiply the time-to-kill by x20 since most people have the aim of a stale potato and the tactics of a toddler.
1. M1014 super (Autoshotgun)
At the top of this list is the autoshotgun, sporting an impressive maximum DPS of 784. This means that a full team of autoshotguns can kill an Expert difficulty tank in at least 2.55 seconds before anyone even has to reload.
The main drawback of this powerhouse is the fact that achieving the maximum damage output requires getting up close and personal, which can easily lead to mistakes or lots of friendly fire.
2. Spas-12 (combat shotgun)
Both Tier 2 shotguns are extremely similar, with the combat shotgun dealing only minimally less DPS than the autoshotgun.
With a top output of 776 DPS, 4 survivors using the combat shotgun and landing every pellet will be able to kill an Expert difficulty tank in at least 2.58 seconds before anyone has to reload.
However, just like the autoshotgun, players need to get close to the tank in order to deal the most damage possible.
3. Ak47
The ak47 is the best weapon in L4D2, having an incredibly high DPS of 435 and no significant downside, other than having to learn how to use it properly.
This means that 4 players can kill an Expert difficulty tank in at least 4.60 seconds from a safe distance without anyone having to reload.
It takes a little bit longer than the autoshotguns, but is far more reliable due to it's range, as well as being the best general-purpose weapon in the game, ranking near the top in every category. All you need to do is learn to manage it's accuracy, and it's one of the strongest weapons in the game for every scenario. The ak47 is the most reliable (and safest) choice you can make.
4. SG552
Despite having stats similar to the M16, the Sg552 boosts a slightly higher maximum damage output at 393 DPS.
This means that a team of 4 players can kill an Expert difficulty tank in at least 5.09 seconds, as long as none of the players scope in.
If all survivors are using the scope for some reason, the time to kill an Expert difficulty tank drops to at least 8.20 seconds due to the slower firerate.
5. Mac-10 (SIlenced SMG)
Despite being a Tier 1 weapon, the Silenced SMG is a surprisingly strong weapon that holds up well even against Tier 2 weapons.
With a top DPS of 379, 4 players with Mac-10s can kill an Expert difficulty tank in at least 5.28 seconds.
6. M16
Being the insecure big brother of the Mac-10, the M16 has a top DPS of 377, which means that a full team of survivors using M16s can kill an Expert difficulty tank in at least 5.31 seconds.
7. Military Sniper/Hunting Rifle
The military sniper and Hunting rifle both have the same damage per shot at 90, and therefore the same maximum DPS of 377.
This allows a full team of snipers to kill an Expert difficulty tank in at least 5.31 seconds.
If they're using the Military sniper specifically, this can be done without anyone needing to reload.
8. Dual pistols
As our first sidearm on the list, the dual pistols sport a considerable maximum of 360 DPS.
This allows a full team of survivors using dual pistols to kill an Expert difficulty tank in at least 5.56 seconds.
9. Scar (Desert Rifle)
As the last of the rifles, the Scar offers a maximum of 359 DPS, making it the weakest Rifle in terms of highest possible damage output.
This means that a full team of SCAR users will be able to kill a tank in at least 5.57 seconds.
10. Mp5
As the middle-ground of the SMGs, the mp5 has a top DPS of 320, which means that a full team of mp5's can shred a tank in at least 6.25 seconds.
11. Uzi (SMG)
With a top DPS of 303, a full team of survivors can kill an Expert difficulty tank in at least 6.6 seconds with uzis.
12. Pump shotgun
Similar to the Tier 2 shotguns, the Tier 1 shotguns are very similar to one another. The pump shotgun sports a lowly 278 DPS at best, allowing a full team of pump shotgun users to kill an Expert difficulty tank in at least 7.19 seconds.
Just like the other shotguns, the Pump shotgun suffers from having to get up close to the tank to deal full damage to it.
13. Chrome shotgun
The chrome shotgun sits ever so slightly lower with a maximum of 276 DPS. This lets a full team of chrome shotgun users kill an Expert difficulty tank in at least 7.25 seconds at best.
Unlike the Pump shotgun, the Chrome has a slightly tighter spread, which means you don't have to get quite as close to the tank in order for all the pellets to land, but in practice this difference is insignificant.
14. Magnum
Our other sidearm, the magnum, is only slightly weaker than the aforementioned shotguns, sporting a top DPS of 267. This allows a full team of magnum users to kill an Expert difficulty tank in at least 7.49 seconds.
15. Pistol
Despite being the starter weapon, and generally the most immediately disregarded weapon, the single pistol is only the 4th slowest way to killing a tank.
With a meager 180 DPS, a full team of single pistol users would need at least 11.11 seconds to kill an Expert difficulty tank.
16. Scout
The bolt snipers both have considerably high damage per shot, but the cycling between each shot drastically lowers their overall DPS, just like with the Tier 1 shotguns and the grenade Launcher.
The Scout tops out at 117 DPS, which allows a full team of Scout users to kill an Expert difficulty tank in at least 17.09 seconds.
17. Awp
The Awp, despite having a slightly higher damage per shot than the Scout, has a slightly lower cycle time, lowering it's top DPS to 108.
This means that a full team of AWP users can kill an Expert difficulty tank in 18.52 seconds under the best possible conditions.
18. Molotov
All the way at the bottom of the list, the molotov sits on a false throne. Dealing only 100 DPS, the molotov has the weakest damage output against tanks of any weapon in the game. This becomes especially apparent when you consider that, in the time it takes to equip and throw a molotov, you could already have dealt upwards of 1000 damage with most other weapons by just shooting instead.
While useful to passively kill tanks with no shooting required, or forcing tanks to commit in Versus, it ultimately is the worst choice for killing a tank quickly, since concentrated gunfire with even the weakest weapons vastly outperforms it's pathetically low damage output. The only time fire damage is useful is in Realism mode, where infected receive damage resistance against guns.
?. The m60
The m60 is essentially an ak47 that doesn't need to reload, with mostly similar stats, and slightly less damage per shot at 50 damage per bullet.
This gives it a grand total of 459 DPS, allowing a full team of m60s to kill an Expert difficulty tank in 4.36 seconds.
However, due to the fact that the M60 doesn't often spawn, it's more likely that only one person finds an M60 for a tank fight, in which case it would take the lonesome Rambo 17.43 seconds to kill an Expert difficulty tank on their own... except that it will run out of ammo before the tank dies.
?. The Grenade Launcher
This one is a bit of a weird one, because the raw numbers will skew how it actually performs. It also rarely spawns like the M60.
It's simpler to say that the grenade launcher has a maximum damage per shot of 1000 on a direct hit, with a 3.3 second reload, you can do the math on this yourself.
There is also the melees, but the general strategy of "run up, swing, back up, repeat" makes them all have a pretty identical DPS value. The only melee which can kill a tank swiftly is the chainsaw, with a DPS of 1000, allowing a full team of chainsaws to kill an Expert difficulty tank in 2 seconds flat, but only if they get up close and personal.
Every other melee will kill a tank in 20 hits on every difficulty, and all of them have the same range, so the only factor that would affect their DPS is their attack speed.
their health pool and ability to one-hit you it daunting at first, but once you learn to abuse a tank's weaknesses they're one of the easiest si to kill.
for the weapons, depending on situation, rifles if you can't afford staying close or someone else has the aggro
shotguns if you got the aggro or are able to juke it around something it can't launch at your face (you can solo a tank this way as well if the tank REALLY focuses you and infected don't slow you down long enough for it to catch up)
m60 if you got it at all
a pipe or propane can stun a tank from what I gathered, definitely helps a lot for melting it's health easly
also melee weapons aren't that bad of an idea if you got luck on your side, since melee can dish out a lot of damage, problem is that you're at range of getting slapped into afterlife if the tank suddenly turns around
