If you click on a staff member then click on their stats tab, you can see they have three stats:

Diagnosis skill, treatment skill and speed.

That tells you everything you need to know about a staff member - when they do a diagnostic they use their diagnosis skill, when they do a treatment they use their treatment skill. Room specific skills will modify one or both of those stats while they are in that specific room.

So for example a nurse with two ranks of the +20% skill for wards will get +40% to diagnosis and +40% to treatment while in a ward.

Room specific skills always add a higher percentage than the general treatment or diagnosis skills, but the general skills work everywhere.
I mean it's pretty self explanatory. GP's office require general practice and diagnosis. All diagnoses rooms require diagnosis skill, such as cardiology, general diagnosis, fluid analysis etc.... Ward requires ward and treatment (optional), same for fracture ward.
Injection room requires injection skill + treatment (for a higher chance of treatment, also optional)
Any treatment rooms require treatment skills, like chromotherapy, the clown clinic, de-lux clinic etc....
The ward doesn't require the ward management skill, but ward management is significantly better than just Treatment or just Diagnosis (as long as you only let the people with ward management work in the wards, and maximize their ward management skill).

There are really only a few skills you *need* to work in certain rooms - Psychiatry, Radiology, Genetics, and Surgery, and maybe Marketing. Janitors need Mechanics to upgrade equipment, too. The other skills just give your people a boost in a specific room (a person with the Pharmacy skill gets a 20% boost to treatment in the Pharmacy Lab as opposed to the 10% boost they get from Treatment. But there's only one level for the Pharmacy skill).

Click on the building and press info.
