Can't get into any of the Colosseums.

I'm in playthrough A, I have the Mysterious letter, but I can't access the elevator that takes you into the colosseum areas. The prompt appears, but it's covered by a red "X". I'm in the middle of the "Weird Machines" quest.

Specifically, I'm trying to get into the Gambler's Colosseum. What character do I need/what NPC do I need to talk to to get into that one? I didn't see any NPC's near the elevator

Only the Forest one required 9S (and the final one at Amusement Park) the rest can be accessed by 2B as early as Route A. Make sure you have the DLC installed because it's part of the DLC.

I have the DLC, and AFAIK, I meet all the requirements for getting in (having the Mysterious Letter, being at least level 25). But I just can't access the arenas. The activation prompt appears, but it's covered by an "X", and nothing happens when I try to activate the doors.

Your main quest stage prevents you from entering arenas. Last chance is before flooded city (you can get to the area before main quest leads you to it).

So now, the only two ways to get there is to either complete this playthrough and try again as 9S, or start a new game?

Its unlikely you'll be able to finish all challenges inside arenas during route A without a lot of needless grinding.
What you are probably supposed to do is to just keep playing, maybe visit arenas a couple of times to collect easier rewards. Then after you finish route C you unlock chapter select. With that you can access arenas as 2B if you want. With all XP and gear from all 3 routes (all playable characters share progression, its just skin and moveset swap basically).
