First of all - keep trying! Uppercuts are super useful punches. I find when I do land an uppercut clean on the chin, it doesn't even have to be hard to be a knockdown.
Second, you need the opponent to open or lower their guard to make space for an uppercut to sneak through. Hooks to the head work well, as do body punches. If you land a couple of hooks to the head, the opponent's guard will open and give you a chance for the uppercut. Harder still, draw out a punch from your opponent by leaning your head in, then slip/pull and counter with the uppercut. This is MUCH harder to do.
Third, I find the distance for the uppercut is really difficult. I either hit the chest (not so terrible), bang into an elbow or shoulder with no damage (not good), or worst, miss completely and then get clobbered with a counter.

Infighting feels (to me) less realistic in TOTF, so I find I need to throw longer range uppercuts than I would IRL boxing. I've had much less success with footwork/head movement driven uppercuts a-la Tyson. https://www.youtube.com/watch?v=SGNV2RKcFFI

@Ian - NOTHING here is a criticism of the game. To the contrary, I'm amazed at how realistic you have made this with the tools available. I can totally understand why realistic infighting is really hard for VR and the methods I describe are above are exactly what I would teach someone - open the guard with hooks and body punches, then come back with uppercuts and straight punches down the middle. Boxing 101 - they can't block everything.
Maybe there could be some game options such as to reduce the opponent's glove size or to space out their hands more when defending, to make uppercuts easier. It wouldn't need to make the game much easier as there could be a risk of counter-attacks when uppercutting.
Then again I play with small gloves to replicate 10-12oz gloves irl.
Practice on Luis as he is smaller and tends to get inside more. Also more aggressive. This will help you find your uppercut range especially if you want to go between fighters too. Once you find your range, try on Reverend, then Artist.

Start streaming and put the in-game window to camera/ringside view on the small window while with mirror window stream you stream the 1st person view. After the live stream (at least in Twitch) you can watch the vod as a replay. I know it's a big workaround but it's something. Although i think there's also a only vodding option for non streamed gameplay recording in Obs/slobs.
I started doing this, and it has been extremely helpful.
