Room Efficiency?
What is this and how do I make it better?

Make sure the room is enclosed with walls and a door for any opening. You can make doors with the tinkerer's room (check the almanac) once the tech is available in campaign missions.

Additionally each and every room needs a specific size in order to be as much effiecient as possible in addition to @odin87654321's post for sure ;)
So e.g. when building a workshop and include a working unit in there, but there are only a few free tiles available, which will be the space for your created toolboxes, it might no work out well, as too less storage is available and always be aware that the working unit needs some space as well as a snot or other horde unit in this specific room also needs some space to work properly.

Indeed. Some buildings work better in different formats (if your room needs 3 x 3 tiles for a production unit, you want to build at least 3 x 6 to accomodate 2 machines and a bunch of free tiles to have space for whatever the machines will produce). The gobbler farm produces chickens only on inner tiles (i.e. if you have a 3 x 3 farm, only the tile in the centre will actually produce delicious bird meat) and so on.

Experiment with the room dimensions to see what works best for you. Keep in mind that not all the efficiency things are actually shown by the room: you want to make sure to have your rooms set up to reduce walking times between them, for example.

You'll want your sleeping room away from where the heros enter so that almost dead monsters don't try to heal up where they will be attacked regularly. You'll want your food farm somewhere near the sleeping room, but also close to any room that regularly gets worked. Your treasure rooms should be as close to the good gold veins as possible (best if built around them, where possible) while also being close to where your monsters spend their time so they can quickly and efficiently grab their pay on payday.

But how do you know what size it needs to be to be efficient? Or am I just not seeing that?

There is no such thing, efficency is just -10% per hole in your room, so without a door You can get max 90% efficency.

Generally, I find that making my rooms 6 by 6 squares is a good default. It's not the most efficient, but it allows quick design without too much thought and the rooms are interchangeable. I build a 4 by 4 treasure room around any gold veins. Temples I make 5 by 5, and I make big mana rooms and big training rooms, and also big libraries. I try to have at least six manequins in the training rooms and 6 research pods in the libraries. With a bit of expansion, it allows you to finish any mission really.

To your original question, some rooms (gobbler farms, training rooms) get efficiency upgrades through the tech trees.
