If it happens again, here some things that might work for Tropico 6:
1. Try to see whether https://mod.io/g/tropico-6 is up and working. Maybe its a web-server issue. If so, nothing you can do but wait.. ;(
2. Try deleting your folder "%LOCALAPPDATA%\mod.io" where mod.io stores the login data and some settings. This might especially help when you have problems logging in.
3. Delete the folder %USERPROFILE%\Documents\My Games\Tropico6\Saved\Paks which contains the downloaded mods. Be sure to ONLY delete the "Paks" folder - your savegames are right next to it. ;). That might expecially help when some mods seem to be stuck on install or uninstall. Then go back into the game and click "Check for Updates" in the mods dialog

So by removing all mods the only difference that makes is that the mods is gone. You will still have problems with http requests.
Tried it myself, so now all my mods are gone :( and ofcourse the useal "http request failed" comes up.
