You can also use certain spells as enchantments. Like soul trap. If you do then this will apply soul trap on the enemy you're fighting, and if you kill the enemy within the allotted time... woosh: you grab their soul.
Some perks need a minimum skill level to be added.
Soul tap is a perk, spell, and weapon enchantment as the others have said, all three result in the same effect of trapping the souls of defeated enemies into soul gems.
The conjuration perk, only applies to weapons created by the "Bound XXXXX" spells, where the X's are the weapon type. And requires a decently leveled conjuration skill to get.
The spell can be purchased from a few places, it is possible to get it in Whiterun, also you can get it in Winterhold too.
The weapon enchant you will have to wait until you find one. Though once find one, odds are can disenchant it so can place it onto a weapon of choice.
Enchanted weapons have their own individual supply of magical power, which can be recharged through the use of soul gems.
The reason you are unable to add the perk points to the skill you want, is perks have skill level requirements and your skill isn't high enough yet. You gain 1 perk each time you level up, but you need to meet the perk's skill requirement in order to spend a perk point on it. Thankfully, you can store up perk points so you can use them when you do meet the requirements. Also on the multi-perk point perks, such as extra 1h damage, each tier of the perk has a different skill requirement.

Soul trap is a SPELL under Conjuration. There is a perk that adds it to bound weapons, but Soul Trap itself is a spell.
It is also an enchantment that can be added to weapons.
No reason for it not to be both.
Any skill confusion you might have is because both the Conjuration and Enchanting skill trees have perks that're RELATED to this effect. Conjuration has the "Soul Stealer" effect, which gives the Soul Trap effect to all your Bound Weapons. For Enchanting, it's "Soul Siphon", which automatically traps a small portion of the target's soul when you hit a creature with a death-blow. However, there is no actual skill nor perk called "Soul Trap", only the two similarly-named perks that I just described.
