Glowing Mushrooms are in MANY caves along the walls.

And yes, you need to fish around with your cursor to hit the activator in several cases.
The spriggan sap will work with the blisterwort to get your your initial upgrade. Then you've got a little bit of time to find your glowing mushrooms (earliest in the main quest line, I believe, is in Bleak Falls Barrow, and plant them for future supply.
You can use glowing mushroom as others have pointed out with blisterwort for a smithing potion, if you want an added bonus, add nightshade to it, gives you smithing and destruction bonus.
Pretty sure you can use sabre cat tooth with glowing mushroom for a smithing bonus as well.
