The Tinkerer is basically just gun-guy galore where you can mod the hell out of all your guns.
The Disquieted has the ability to teleport to people when wielding a melee weapon and targetting them, as well as the ability to perform a finisher move on somebody with said melee weapon at the cost of breaking it. You can't remove his mask however.
The Massive gets bigger and bigger as you level up, and you can only upgrade his strength & endurance.
I have yet to play Tinkerer or The Massive, but so far I'm really enjoying The Disquieted.
These may not be in the game in it's current state, but that doesn't mean they won't be added post launch. Plus, the devs are working closely with modders, and making sure that the game post-launch will become very moddable. This means even if the devs don't add it, I'm sure some modders will. I heavily recommend this game and have hundreds of hours in it and have played since early alpha.
