It could be a pathfinding issue which can be really tricky to spot and sort out. I'd eliminate everything but the bed just to try that out, and ensure there are two full spaces around both sides of the bed and its foot, just to see if that helped.
The part that makes me think it might be a pathfinding issue is that the bed works if it's on the first floor - that means there's something about it being on the second floor that is making one side of the bed inaccessible. The big red X over the person means "that person's already sleeping there" - that is, your Sim isn't recognizing the other side of the bed as somewhere they can get to. I think.
The bad news - looks like it's a glitch that's still present in Sims 4. :-|
And the weird part about the person they think about is sometimes they are plumbots who are not in the bed. Since this post, it happened in another room, and the only fix I have found is to clone the bed, sell the current one, and replace it with the cloned bed. The cloned beds seem to always work, even in the original spots. So now I am forced to use the money cheat to compensate, as lossing money over this seems very broken.
The Sim could pathfind past that side when it was empty, but could not when someone was sleeping in it, or making it (or queued up on their way to make it) and so on. That's why I suggested clearing space around the bed, though it's hard to say what will really help. If you can, post a screenshot of the bedroom (with grid on, if possible) and some of us might be able to try figure it out better.

The only other thing I can think of, is using testingCheatsEnabled or MasterController (from NRaas) to reset the beds and seeing if that works.
