As for the turret, there are a few options, some of which you might already know. First of all, you can set the turret to either aim at your current target or at your mouse position under the game options. Then you can also choose a control scheme of keys/mouse+keys/mouse only.
I'm afraid there's no separate turret/front gun firing in the game yet. We thought about it for some time, but ended up deciding against it, as it would have added even more keys and make the controls more complicated than they already were.
For the controls you are asking about, I think the closest we can currently get is this: Set the turret to fire at the mouse position, and the control scheme to "mouse only". By deactivating "Heading" (the furthest away button of the three in front of your ship) you will be able to turn the ship normally with the WASD keys, and can fire both your main weapons with the mouse.
I hope that helps you to some extent. If all you really want is a main turret-type gun controlled with the mouse, then you can also sell your front gun and use the method I described above to get full mouse control (aiming and firing) for the turret.
A more complicated control scheme wouldn't necessarily be a bad thing, you would not have to have it as the default scheme and it could even be labelled "Ädvanced Controls". Maybe, Keys to control the ship, space bar to turn mouse control of the turret on and off (switch between mouse target mode and weapon mode, so the weapon doesn't fire when you click on something to target it), left click fire turret, middle mouse button fire front gun, just an idea, I'm sure you have plenty to do already.

Edit: The only problem with the control scheme you suggested is that when I'm mining, targeting asteroids shoots the weapon, so it's not really usable.
Edit 2: Maybe a simpler way to deal with turrets would be to be able to manually target ships and debris etc by clicking on them with the mouse much like you target asteroids and then you can set the turret to the Target/Front option. (At the moment I can't seem to find a cancel target button (that could be just me missing it though) so the Target/Front option is very awkward to use now)
If we add separate turret controls, that would need to include a proper tutorial and in-game information on how to use it, which could end up being too complicated for most users. We'll think about it some more, but I really can't promise anything in that area, sorry.
About the un-targeting: There is a "X" checkbox at the top right next to the current target display, which you can click to remove the current target. FYI there's still the bug that fires your guns when you click on UI boxes, but we're planning on fixing that for the next update.
I can't help thinking that a lot of the fans of this game/genre would be pretty used to byzantine control systems already, compared to some of the 3d space sims the controls in 3030 are pretty simple. But obviously you know what is best for your game, I appreciate all your hard work and am very pleased to have found such a gem.

Definitely use the turret instead of main guns and leave it on the setting to fire at target. It will track and fire at enemies as you maneuver around them without having to turn and face them.
Old thread, but since it got unburied, I feel like I should mention that I think what cuts weapon damage in half is having two weapons at once (front and turret mount). Having the turret by itself doesn't cause a damage penalty.
